TNF-a Suppresses a-Smooth Muscle Actin Expression in Human Dermal Fibroblasts: An Implication for Abnormal Wound Healing

چکیده
Abnormal wound healing encompasses a wide spectrum, from chronic wounds to hypertrophic scars. Both conditions are associated with an abnormal cytokine profile in the wound bed. In this study, we sought to understand the dynamic relationships between myofibroblast differentiation and mechanical performance of the collagen matrix under tissue growth factor–b (TGF-b) and tumor necrosis factor–a (TNF-a) stimulation. We found TGF-b increased a-smooth muscle actin (a-SMA) and TNF-a alone decreased the basal a-SMA expression. When TGF-b1 and TNF-a were both added, the a-SMA expression was suppressed below the baseline. Real-time PCR showed that TNF-a suppresses TGF-b1-induced myofibroblast (fibroproliferative) phenotypic genes, for example, a-SMA, collagen type 1A, and fibronectin at the mRNA level. TNF-a suppresses TGF-b1-induced gene expression by affecting its mRNA stability. Our results further showed that TNF-a inhibits TGF-b1-induced Smad3 phosphorylation via Jun N-terminal kinase signaling. Mechanical testing showed that TNF-a decreases the stiffness and contraction of the lattices after 5 days in culture. We proposed that changes in a-SMA, collagen, and fibronectin expression result in decreased contraction and stiffness of collagen matrices. Therefore, the balance of cytokines in a wound defines the mechanical properties of the extracellular matrix and optimal wound healing.

Loss of PPARγ expression by fibroblasts enhances dermal wound closure
BACKGROUND Peroxisome proliferator-activated receptor (PPAR)γ may be a key regulator of connective tissue deposition and remodeling in vivo. PPARγ expression is reduced in dermal fibroblasts isolated from fibrotic areas of scleroderma patients; PPARγ agonists suppress the persistent fibrotic phenotype of this cell type. Previously, we showed that loss of PPARγ expression in fibroblasts resulted...

Wound healing impairment is a well-documented phenomenon in clinical and experimental diabetes, and in diabetic wound healing impaired fibroblast has been linked to increased levels of tumor necrosis factor-α (TNF-α). A number of signaling pathways including TNF-α/forkhead box O1 (FOXO1) and transforming growth factor-β1 (TGF-β1)/Smads in fibroblasts appear to play a cardinal role in diabetic w...

Previous studies indicate that organ fibroblasts play an important role in wound healing, collagen production, remodeling processes and pathogenesis of progressive heart, lung, renal and hepatic fibrotic diseases. Several studies suggest a possible inhibitory role for adenosine in the regulation of fibroblast proliferation. The effect of adenosine A2 agonists on proliferation and differentiatio...
متن کاملTGF-ß1 Latency Associated Peptide Promotes Remodeling of Healing Cutaneous Wounds in the Rat
Background: The process of wound healing involves integrated events including inflammation, granulation tissue formation, matrix deposition and remodeling. Growth factors play a key role in the process. Among them transforming growth factor-ß1 (TGF-ß1) is known to accelerate tissue repair by promoting the synthesis and deposition of extracellular matrix proteins. However, persistence or overact...
متن کاملTAT-Mediated Acidic Fibroblast Growth Factor Delivery to the Dermis Improves Wound Healing of Deep Skin Tissue in Rat
BACKGROUND The definition of deep tissue injury was derived from multiple clinical cases as "A purple or maroon localized area of discolored intact skin or blood-filled blister due to damage of underlying soft tissue from pressure and/or shear". Acidic fibroblast growth factor (aFGF) significantly improves wound healing under diabetic conditions. However, to date, the therapeutic application of...
